چکیده

Cyanate is undergoing study as a drug to prevent occlusive sickle cell crises. Carbamylation of hemoglobin S increases its oxygen affinity, thereby decreasing its tendency to aggregate at low oxygen tensions. The cell membrane has also been shown to be carbamylated. We studied the effect of carbamylation on deformability of sickle, normal, and stored normal erythrocytes. Deformability was measured as filtration time of erythrocyte suspensions through 3-z polycarbonate filters. Oxygenated carbamylated sickle erythrocytes had a marked shortening of filtration time that could not be explained by changes in morphology. In addition, normal erythrocytes became more filterable with carbamylation. Decreased filterability after storage at 4’ C for 48 hr was returned to normal following carbamylation and incubation with glucose. No correlation was made with changes in either morphology, cell size, or ATP levels. Therefore, it was postulated that carbamylation affected filterability by binding either the cell membrane or cell contents or both.

The effect of centrifugation at various g force levels on rheological properties of rat, dog, pig and human red blood cells.

Laboratory investigations often require centrifugation of blood samples for various erythrocyte tests. Although there is a lack of data about the effect of centrifugation at various g force levels on erythrocyte rheological properties. We aimed to investigate the effect of a 10-minute centrifugation at 500, 1000 or 1500 g at 15°C of rat, dog, pig and human venous (K3-EDTA, 1.5 mg/ml) blood samp...

Effect of anti-spectrin antibody and ATP on deformability of resealed erythrocyte membranes.

Deformability of resealed erythrocyte membranes was measured by using an ektacytometer. Divalent anti-spectrin antibody, but not monovalent anti-spectrin Fab fragments, decreased membrane deformability. Membranes resealed with MgATP were more deformable than those without MgATP. Exogenous alkaline phosphatase, which dephosphorylates spectrin, decreased membrane deformability. These results sugg...

The effect of L-carnosine on erythrocyte deformability and aggregation according to the cell age in young and aged rats.

This study aimed to investigate alterations in hemorheology induced by L-carnosine, an anti- oxidant dipeptide, and to determine their relationship to oxidative stress in density-separated erythrocytes of aged and young rats. 28 male Sprague Dawley rats were divided into 4 groups as aged (Aca), young (Yca) L-carnosine groups (250 mg/kg L-carnosine, i.p.) and aged (As), young (Ys) control groups...

The Functional Significance of the Rho/Rho-Kinase Pathway in Human Erythrocytes

OBJECTIVE Erythrocyte deformability, which can be influenced by various intracellular signaling mechanisms, such as nitric oxide, cAMP, cGMP, and protein kinases, is the most important physiological factor providing the blood flow in microcirculation. However, the functional significance of the Rho/Rho-kinase pathway, which contributes cell shape changes and the reorganization of the actin cyto...

Decreased deformability of erythrocytes from smokers.

The deformability of erythrocytes from smoking and non-smoking human subjects was examined by filtration through 3-mu pores and capillary viscometry of cell suspensions. In both cases, small but significant differences were found between the two groups that are consistent with a reduction in the surface area-to-volume ratio and/or a diminished membrane flexibility in erythrocyte from smokers. A...
